This stain kit is commonly used for staining oocysts of the Cryptosporidium species in foecal smears. It is also useful to confirm the presence of oocysts of Isospora belli and Cyclospora cayetanensis

Results:
Oocysts of Cryptosporidium, parvum: Magenta / Red
Oocysts of Isospora belli: Granular / Red
Oocysts of Cyclospora cayetanensis: Variable Staining (some are acid fast giving a round hole of glassy wrinkled sphere
Protocol:
- Immerse in the Cryptosporidium Fixative for 3 minutes
- Flood the slides with carbol fuchsin Kinyoun for 20 minutes
- Wash well in distilled water
- Differentiate with TB differentiator until preparation is colourless or pale pink (until no more dye runs out), approximately 10 seconds
- Wash well in distilled water
- Counterstain with malachite green for 1 minute
- Wash well in distilled water
- If oil immersion is to be used for visualisation of smears the preparation must be air dried for 20-30 minutes
