This stain kit is used for the detection of Amyloid Protein in tissue sections. The use of alkaline alcoholic solvents has the effect of supressing staining of other tissue components whilst enhancing hydrogen bonding hence improving selectivity for amyloid. This method has stable solutions and has a high degree of selectivity

Kit Contains:
Congo Red in 50% ethanol
Haemalum Mayer
0.2% Potassium Hydroxide in 80% ethanol

Results:
Eosinophils, Amyloid, Elastin,Keratin: Orange/Red
Nuclei: Blue
Protocol:
- Dewax sections, hydrate through alcohols and rinse in tap water
- Place in congo red Solution for 15 minutes
- Differentiate in potassium hydroxide Solution for 3-10 seconds
- Wash in running tap water
- Stain nuclei with haemalum Mayer for 5 minutes
- Wash well in water
- Differentiate in 1% acid alcohol for 2-10 seconds
- Wash well in water
- Blue in Scotts tap water
- Wash well in tap water
- Dehydrate rapidly, clear and mount
